Takahashi K, Yamanaka S. 2006) \u5f8c\u53ef\u8b8a\u6210\u548c\u80da\u80ce\u5e79\u7d30\u80de\u4e00\u6a23\u7684\u591a\u80fd\u5e79\u7d30\u80de\uff0c\u800c\u4e14\u53ef\u907f\u958b\u80da\u80ce\u5e79\u7d30\u80de\u53d6\u81ea\u80da\u80ce\u7684\u502b\u7406\u722d\u8b70\uff0c\u5f88\u6709\u53ef\u80fd\u53d6\u4ee3\u80da\u80ce\u5e79\u7d30\u80de\uff0c\u958b\u555f\u6cbb\u7642\u65b0\u5951\u6a5f\u3002\u57282007\u5e74\u7814\u7a76\u5718\u968a\u53c8\u767c\u73fe\u4eba\u985e\u7e96\u7dad\u7d30\u80de\u4e5f\u53ef\u8b8a\u6210iPScs (Takahashi et al., 2007)\u3002\u540c\u4e00\u5929\uff0cJames Thomson\u7814\u7a76\u5718\u968a\u4e5f\u767c\u73feOct 4, Sox2, Nanog, Lin28\u53ef\u8f49\u8b8a\u4eba\u985e\u9ad4\u7d30\u80de\u8b8a\u6210iPSc (Yu et al., 2007)\u3002<\/p>\n<p style=\"text-indent: 2em\">\u5c71\u4e2d\u4f38\u5f4c\u7562\u696d\u65bc\u795e\u6236\u5927\u5b78\u91ab\u5b78\u9662\uff0c\u672c\u4f86\u7acb\u5fd7\u7576\u6574\u578b\u79d1\u91ab\u5e2b\uff0c\u4f46\u537b\u56e0\u5be6\u7fd2\u6642\u6280\u8853\u4e0d\u4f73\uff0c\u53ea\u597d\u9078\u64c7\u7814\u7a76\u4e00\u9014\u3002\u5728\u7f8e\u570b\u9032\u884c\u535a\u5f8c\u7814\u7a76\u6642\uff0c\u6b63\u7576\u79d1\u5b78\u5bb6\u5011\u56e0\u80da\u80ce\u5e79\u7d30\u80de\u7684\u502b\u7406\u722d\u8b70\u4e0d\u4f11\uff0c\u800c\u555f\u767c\u5c71\u4e2d\u767c\u73feiPScs\uff0c\u5c71\u4e2d\u5728\u5f97\u734e\u5f8c\u5728Cell press\u4e2d\u64b0\u6587\u6709\u95dciPSc\u7684\u904e\u53bb\uff0c\u73fe\u5728\u8207\u672a\u4f86\u5c55\u671b\uff0c\u4eca\u7bc0\u8b6f\u5176\u4e2d\u6587\u7ae0\uff0c\u8207\u5927\u5bb6\u5206\u4eab\u3002<\/p>\n<p>\u555f\u767c\uff1a<br \/>\niPScs\u7684\u767c\u73fe\u4e3b\u8981\u662f\u532f\u96c6\u4e09\u5927\u79d1\u5b78\u7684\u4e3b\u6d41\u800c\u6210 (\u5716\u4e00)<\/p>\n<p><img decoding=\"async\" src=\"http:\/\/wp.kmu.edu.tw\/rd\/files\/2012\/11\/002P01.jpg\" alt=\"\" \/><\/p>\n<p style=\"text-indent: 2em\">\u9996\u5148\u662f\u8907\u88fd\u52d5\u7269\u7684\u89c0\u5ff5\uff1a\u628a\u6210\u9ad4\u7d30\u80de\u6838\u6253\u5165\u53bb\u6838\u7684\u5375\u5167\uff0c\u6210\u529f\u5730\u8907\u88fd\u52d5\u7269\uff0c\u9019\u8868\u793a\u6210\u9ad4\u7d30\u80de\u542b\u6709\u6240\u6709\u9577\u6210\u500b\u9ad4\u7684\u57fa\u56e0\uff0c\u800c\u5375\u542b\u6709\u67d0\u4e9b\u56e0\u5b50\u53ef\u4ee5\u555f\u52d5\u6539\u7de8\u9019\u4e9b\u57fa\u56e0\u3002\u9032\u4e00\u6b65\u5730\u79d1\u5b78\u5bb6\u4e5f\u8a3c\u660e\u80da\u80ce\u5e79\u7d30\u80de\u5167\u4ea6\u542b\u6709\u9019\u4e9b\u6539\u7de8\u7684\u56e0\u5b50\u3002\u7b2c\u4e8c\u500b\u89c0\u5ff5\u662f\u63a7\u5236\u8f49\u9304\u56e0\u5b50\u5373\u80fd\u63a7\u5236\u4e0d\u540c\u80da\u5c64\u6216\u8868\u9ad4\u8868\u73fe\uff0c\u5982\u63a7\u5236\u679c\u8805\u89f8\u89d2\u751f\u6210\u7684Antennapedia\u8f49\u9304\u56e0\u5b50\uff0c\u5982\u679c\u5728\u817f\u90e8\u8868\u73fe\u4e5f\u53ef\u63a7\u5236\u817f\u7684\u9577\u6210\uff0cMyoD\u9019\u500b\u54fa\u4e73\u985e\u8f49\u9304\u56e0\u5b50\uff0c\u53ef\u4ee5\u628a\u7e96\u7dad\u7d30\u80de\u8f49\u8b8a\u6210\u808c\u7d30\u80de\u3002\u7b2c\u4e09\u500b\u89c0\u5ff5\uff0c\u540c\u6a23\u5f88\u91cd\u8981\u7684\u5c31\u662f\u80da\u80ce\u5e79\u7d30\u80de\u7684\u7814\u7a76\uff0c\u79d1\u5b78\u5bb6\u5011\u7814\u7a76\u51fa\u80fd\u9577\u671f\u57f9\u990a\u80da\u80ce\u5e79\u7d30\u80de\u7684\u56e0\u5b50\uff0c\u50cfLIF, bFGF\u80fd\u7dad\u6301\u80da\u80ce\u5e79\u7d30\u80de\u7684\u5168\u80fd\u6027\u3002<\/p>\n<p style=\"text-indent: 2em\">\u9019\u4e9b\u904e\u53bb\u7684\u91cd\u8981\u89c0\u5ff5\uff0c\u555f\u767c\u4e86Yamanaka\u7b49\u4eba\uff0c\u5927\u81bd\u5047\u8a2d\uff0c\u5728\u5375\u5b50\u6216\u80da\u80ce\u5e79\u7d30\u80de\u5b58\u5728\u6709\u67d0\u4e9b\u56e0\u5b50\uff0c\u53ef\u4ee5\u4f7f\u9ad4\u7d30\u80de\u8fd4\u56de\u80da\u80ce\u6642\u671f\u72c0\u614b\uff0c\u800c\u4e14\u914d\u5408\u7dad\u6301\u80da\u80ce\u5e79\u7d30\u80de\u7684\u57f9\u990a\u6db2\uff0c\u4ed6\u5011\u7d42\u65bc\u627e\u5230\u56db\u500b\u56e0\u5b50\uff0c\u53ef\u4ee5\u7522\u751fiPScs\u3002<\/p>\n<p style=\"text-indent: 2em\">iPSc\u6280\u8853\u7684\u91cd\u8981\u6027\u70ba\u5b83\u662f\u7c21\u55ae\u4e14\u518d\u73fe\u6027\u9ad8\u7684\u6280\u8853\uff0c\u4f46\u5b83\u7684\u6548\u80fd\u8f03\u4f4e\uff0c\u4e00\u822c\u4f86\u8aaa\uff0c\u5c11\u65bc1%\u7684\u7e96\u7dad\u7d30\u80de\u53ef\u8f49\u6210iPScs\uff0c\u9019\u5176\u4e2d\u5c1a\u5b58\u5728\u4e00\u4e9b\u554f\u984c\u6c92\u6709\u88ab\u89e3\u6c7a\u3002\u5728\u9f20\u7814\u7a76\u4e2d\u4f7f\u7528retroviruses\u6216lentiviruses\u4f86\u8f49\u67d3\uff0c\u5728\u4eba\u7684\u7814\u7a76\u9700\u907f\u514d\uff0c\u6240\u4ee5\u5f8c\u7e8c\u7814\u7a76\uff0c\u5305\u62ec\u4f7f\u7528plasmid (Okita et al., 2011; Okita etal., 2008)\uff0cSendai virus (Fusaki et al., 2009)\uff0cadenovirus (Stadtfeld et al., 2008)\uff0c\u5408\u6210\u7684RNAs (Warren et al., 2010) \u53ca\u86cb\u767d (Kim et al., 2009)\u3002\u5176\u4e2d\uff0cPlasmid\u53caSendai Virus\u6700\u5ee3\u70ba\u76ee\u524d\u5be6\u9a57\u5ba4\u4f7f\u7528\u3002<\/p>\n<p style=\"text-indent: 2em\">\u767c\u73feiPScs\u6280\u8853\u5f8c\uff0c\u79d1\u5b78\u5bb6\u958b\u59cb\u7814\u7a76iPScs\u7684\u518d\u751f\u91ab\u5b78\u61c9\u7528\uff08\u5716\u4e8c\uff09\u5982\u6cbb\u7642Parkinson\u2019s\u00a0 disease (kribs et al., 2011)\uff0c\u8840\u5c0f\u677f\u7f3a\u4e4f (Takayama et al., 2010)\uff0c\u810a\u67f1\u640d\u50b7 (Nori et al., 2011; \u00a0\u00a0 \u00a0Tsuji et al., 2010) \u53ca\u9ec3\u6591\u75c5\u8b8a (Okamoto and Takahashi 2011)\u3002\u53e6\u5916\uff0c\u7531\u75c5\u4eba\u7d30\u80de\u4f86\u7684iPScs\u53ef\u7576\u4f5c\u85e5\u7269\u7be9\u9078\u5e73\u53f0\uff0c\u50cfParkinsons disease (Devine et al., 2011)\uff0cAlzheimer\u2019s disease (Israel et al., 2012) \u53ca\u7cbe\u795e\u5206\u88c2\u75c7 (Brennand et al., 2011)\u3002\u7531\u9ad4\u7d30\u80de\u4f86\u7684iPScs\uff0c\u5982\u5fc3\u808c\u53ca\u809d\u7d30\u80de\uff0c\u53ef\u7576\u6210\u6bd2\u7406\u5b78\u6e2c\u8a66\u3002<\/p>\n<p style=\"text-indent: 2em\">\u9664\u4eba\u9ad4\u518d\u751f\u91ab\u5b78\u61c9\u7528\u5916\uff0ciPScs\u53ef\u61c9\u7528\u65bc\u52d5\u7269\u751f\u7269\u6280\u8853\u3002\u7334\u3001\u8c6c\u3001\u53ca\u72aciPScs\u53ef\u7528\u65bc\u75be\u75c5\u52d5\u7269\u7684\u5efa\u7acb\u3002\u4ee4\u4eba\u6700\u9707\u596e\u7684\u5831\u5c0e\u70ba (Kobayashi et al., 2010)\uff0c\u7f3a\u4e4f\u80f0\u81df\u7684\u57fa\u56e0\u4e0d\u5168\u5c0f\u9f20\uff0c\u4f7f\u7528\u5927\u9f20iPScs\u6253\u5165\u6b64\u5c0f\u9f20\u56ca\u80da\u7d30\u80de\u4e2d\uff0c\u5728\u5c0f\u9f20\u4e2d\u53ef\u7522\u751f\u5927\u9f20\u80f0\u81df\uff0c\u61c9\u7528\u6b64\u6280\u8853\uff0c\u53ef\u80fd\u5728\u52d5\u7269\u4e2d\u88fd\u9020\u4eba\u985e\u5668\u5b98\uff0c\u7528\u65bc\u79fb\u690d\u4e4b\u7528\u3002\u800c\u6709\u4e9b\u7814\u7a76\u5247\u6307\u51fa\uff0c\u5229\u7528iPScs\u7d30\u80de\u53ef\u4f7f\u7e96\u7dad\u7d30\u80de\u8f49\u8b8a\u6210\u795e\u7d93\u7d30\u80de\uff0c\u809d\u7d30\u80de\uff0c\u5fc3\u808c\u7d30\u80de\u53ca\u9020\u8840\u5e79\u7d30\u80de (Qian et al., 2012)\u3002<\/p>\n<p><img decoding=\"async\" src=\"http:\/\/wp.kmu.edu.tw\/rd\/files\/2012\/11\/002P02.jpg\" alt=\"\" \/><\/p>\n<p style=\"text-indent: 2em\">iPScs\u6280\u8853\u5df2\u6f38\u9a45\u6210\u719f\uff0c\u66f4\u9032\u4e00\u6b65\u7684\u61c9\u7528\uff0c\u5982\u5e79\u7d30\u80de\u6cbb\u7642\uff0c\u4f46\u662f\u5982\u4f55\u9078\u64c7\u6700\u4f73\u7684iPScs clones\u4ecd\u9700\u66f4\u591a\u7684\u7814\u7a76\u3002\u5229\u7528\u57fa\u56e0\u9ad4 (genome) \u53ca\u5916\u57fa\u56e0\u9ad4\u5b78 (epigenome) \u53ef\u80fd\u53ef\u9054\u00a0 \u5230\u6700\u4f73\u9078\u64c7\uff0c\u800c\u5229\u7528\u540c\u8ceaHLA\u7684\u6350\u8d08\u8005\u7522\u751fiPScs\u53ef\u964d\u4f4e\u6392\u65a5\u7684\u554f\u984c (Okita et al., 2011)\u3002\u6709\u8da3\u7684\u662f\uff0c\u53e6\u4e00\u500b\u5f97\u734e\u8005John Gurdon\uff0c\u56e0\u767c\u73fe\u301dclone\u301e\u6280\u8853\u800c\u5f97\u734e\uff0c\u572815\u6b72\u6642\uff0c\u5728\u751f\u7269\u9019\u9580\u8ab2\uff0c\u537b\u662f\u5168\u73ed250\u4eba\u4e2d\u7684\u5012\u6578\u7b2c\u4e00\u540d\uff0c\u8001\u5e2b\u5c0d\u4ed6\u7684\u8a55\u8a9e\u66f4\u662f\u6158\u4e0d\u5fcd\u7779\uff0c\u4f46\u5169\u4f4d\u79d1\u5b78\u5bb6\u90fd\u57f7\u8457\u4e8e\u81ea\u5df1\u7684\u7814\u7a76\uff0c\u4e0d\u754f\u4eba\u751f\u4e2d\u7684\u4efb\u4f55\u632b\u6557\uff0c\u7d42\u7a76\u8b93\u6614\u65e5\u5632\u7b11\u4ed6\u5011\u7684\u540c\u5115\u6216\u8001\u5e2b\u5011\u8dcc\u7834\u773c\u93e1\u3002<\/p>\n<p style=\"text-align: right\"><a href=\"http:\/\/wp.kmu.edu.tw\/rd\/archives\/206\">\u56de\u7b2c0002\u671f\u4e3b\u9801<\/a><\/p>\n","protected":false},"excerpt":{"rendered":"<p>\u56de\u7b2c0002\u671f\u4e3b\u9801 \u4e00\u3001\u7814\u7a76\u65b0\u77e5 2012\u5e74\u8afe\u8c9d\u723e\u751f\u7406\u91ab\u5b78\u734e\u5f97\u4e3b\u5c71\u4e2d\u4f38\u5f4c\u6559\u6388\u8ad6\u6587 &hellip;
