August 2015, Volume 6:8332.<\/p>\n<p>Methionine restriction extends the lifespan of various model organisms.<br \/>\nLimiting S-adenosylmethionine (SAM) synthesis, the first metabolic reaction<br \/>\nof dietary methionine, extends longevity in Caenorhabditis elegans but<br \/>\naccelerates pathology in mammals. Here, we show that, as an alternative to<br \/>\ninhibiting SAM synthesis, enhancement of SAM catabolism by glycine N-<br \/>\nmethyltransferase (Gnmt) extends the lifespan in Drosophila. Gnmt strongly<br \/>\nbuffers systemic SAM levels by producing sarcosine in either high-methionine<br \/>\nor low-sams conditions. During ageing, systemic SAM levels in flies are<br \/>\nincreased. Gnmt is transcriptionally induced in a dFoxO-dependent manner;<br \/>\nhowever, this is insufficient to suppress SAM elevation completely in old<br \/>\nflies. Overexpression of gnmt suppresses this age-dependent SAM increase and<br \/>\nextends longevity. Pro-longevity regimens, such as dietary restriction or<br \/>\nreduced insulin signalling, attenuate the age-dependent SAM increase, and<br \/>\nrely at least partially on Gnmt function to exert their lifespan-extending<br \/>\neffect in Drosophila.
